The Problem with Human Intestinal Organoids
Human intestinal organoids have become a workhorse model system for understanding normal intestinal development and mechanisms of colon cancer growth. Organoids, derived from intestinal stem or cancer cells, grow in three dimensions with odd shapes and inconsistent cell surface areas resulting in difficultly building robust and reproducible assays.
RepliGut® Systems
RepliGut® Systems are a series of 2-dimensional human primary intestinal epithelial cell model systems that recreate the gastrointestinal cell barrier in standard culture well formats. Using the same starting cellular material as organoids, these models deliver more reproducible assay results with consistent culture timelines across a range of pharmacological applications including inflammation, toxicity, bioavailability, and barrier function.

How we do it:
Step 1
We source human intestinal epithelial stem cells from donor tissue while upholding ethical standards
Step 2
By expanding these stem cells, we can create a cryopreserved biobank that allows for multiple studies using the same donor.
Step 3
Our biobanked cells are carefully thawed and then directly seeded onto a semi-permeable membrane coated with a biomimetic scaffold.

Human intestinal cell barrier model is organized as a single polarized layer of differentiated epithelial cells with tight junctions.
1. We source human intestinal epithelial stem cells from donor tissue while upholding ethical standards.
2. By expanding these stem cells, we can create a cryopreserved biobank that allows for multiple studies using the same donor.
3. Our biobanked cells are carefully thawed and then directly seeded onto a semi-permeable membrane coated with a biomimetic scaffold.

Kits
RepliGut® Kits are available in both 12-well and 96-well transwell plate formats to accomodate scientist's needs. Kits are sold complete with the cells, plates, and media needed to culture this unique human intestinal epithelial cell model in your own facility. Technical support is available.
Services
All service projects are led by scientists with extensive experience in advanced cell culture systems. Assays are customizable based on your needs and can be run using standard or customized cell culture conditions.
Multiple Donor Biobank
Our biobank consists of multiple intestinal regions from multiple donors, allowing researchers to study colon or small intestine-specific biology in demographically diverse backgrounds (e.g. age, sex, blood type). Our cryopreserved biobank enables the ability to work with the same donor as your project progresses
